Abstract
Latar belakang : Labu siam (Sechium edule) memiliki kaya akan kandungan antioksidan seperti alkaloid, flavonoid, tannin, saponin, flavonoid, dan golongan vitamin (C,E,A). Penelitian ini bertujuan untuk mengetahui perbandingan gambaran histopatologi paru tikus putih (Rattus norvegicus) jantan galur Sprague dawley yang diinduksi paraquat dan diberi ekstrak labu siam (Sechium edule).
Metode : Penelitian ini merupakan penelitian eksperimental post test only control group design. Subjek penelitian menggunakan bahan biologis tersimpan berupa 20 sampel organ paru tikus putih (Rattus norvegicus) yang sudah diberi pelakuan pada penelitian sebelumnya yang terbagi menjadi 4 kelompok, yaitu kelompok kontrol negatif (P1; diberikan akuades), kelompok kontrol positif (P2; diberikan paraquat 10 mg/kgBB [single dose intraperitoneal]), kelompok perlakuan I (P3; diberikan paraquat 10 mg/kgBB [single dose intraperitoneal] dan vitamin C dosis 0,075 g/kgBB), dan kelompok perlakuan II (P4; diberikan paraquat 10 mg/kgBB [single dose intraperitoneal] dan ekstrak buah labu siam [Sechium edule] dosis 0,75 g/kgBB). Semua sampel bahan biologis tersimpan dilakukan pembuatan preparat paru. Penilaian keadaan paru dilakukan menggunakan kriteria American Thoracic Society dengan bantuan aplikasi ImageJ. Data dianalisis dengan uji Kruskal-Wallis dan uji Mann-Whitney.
Hasil penelitian : Hasil uji Kruskal-Wallis menunjukan terdapat perbedaan skor kerusakan paru yang bermakna antar kelompok dengan nilai p yaitu 0,03 (p<0,005). Pada penilaian skor kerusakan kelompok P4 memiliki skor kerusakan lebih rendah dibanding P2 dan P3. Hasil uji Mann Whitney juga menunjukkan perbedaan yang tidak bermakna kelompok P4 dan P1 dengan nilai p = 0,052 (p<0,005).
Kesimpulan : Pemberian ekstrak buah labu siam dosis 0,75 g/kgBB mampu memperbaiki kerusakan paru tikus putih yang diinduksi paraquat dengan dinilai menggunakan kriteria American Thoracic Society dengan bantuan aplikasi ImageJ. Pemberian ekstrak labu siam mampu memperbaiki kerusakan paru tikus putih lebih baik daripada vitamin C berdasarkan nilai rata-rata skor kerusakan paru.
