Abstract
Pada budidaya udang intensif pakan merupakan komponen utama dan membutuhkan 50-60% dari biaya produksi. Pemberian pakan yang berlebih dalam budidaya intensif mengakibatkan munculnya limbah organik pakan yang secara kimiawi membentuk senyawa amoniak yang bersifat toksik terhadap udang yang dibudidayakan. Senyawa amoniak dapat mengganggu pertumbuhan udang dan bahkan membunuh udang secara masal sehingga tambak mengalami kegagalan panen. Komponen utama dalam limbah organik sisa pakan adalah protein, sehingga untuk mengatasi permasalahan limbah ini adalah dengan cara melakukan remediasi proteinnya. Proeses remediasi protein limbah pakan dapat dilakukan oleh bakteri proteolitik. Bakteri Bacillus Firmus dari simbion sponge Chalinula Pseudomolitba merupakan salah satu bakteri proteolitik yang menghasilkan enzim protease ekstraseluler. Bakteri Bacillus Firmus memiliki potensi untuk dikembangkan menjadi probiotik dalam budidaya udang. Dalam upaya tersebut diperlukan informasi optimasi konsentrasi co-substrat glukosa sebagai sumber C dan amonium nitrat sebagai sumber N dalam media kultur bakteri Bacillus firmus untuk produksi biomassa bakteri. Tujuan dari penelitian ini adalah melakukan optimasi konsentrasi glukosa dan amonium nitrat media kultur Bakteri Bacillus Firmus terhadap pertumbuahan dan aktivitas protease ekstraseluler. Penelitian menggunakan metode eksperimen aboratoris. Penelitian telah berhasil melakukan uji optimasi konsentrasi glukosa dan amonium nitrat media kultur. Hasil penelitian menunjukkan bahwa bakteri Bacillus firmus memiliki laju pertumbuah dan aktivitas protease terbaik pada penambahan co-subtrat glukosa sebagai sumber karbon pada konsentrasi 1% yaitu dengan nilai sebesar 0,145 c dan 23,960 c IU/ml. Selain itu juga, dengan penambahan co-subtrat amonium nitrat sebagai sumber nitrogen pada konsentrasi 0,05% sebesar 0,157 c dan 27,514 c IU/ml.
